With my transfer case remove can i still start the motor and shift through the gears on a transmission

You cannot shift through gears if it is an automatic trans. Without the drive shaft to stop the output shaft from turning, you will damage the transmission. Also with the driveshaft removed you could lose transmission fluid with the engine running.
