I have a posting for a 2005 Highlander and did this with Craigslist. I got a few
views with you but now I am getting many requests from Craigslist and it is
freaking me out a bit. They all want to pay cash at the full price. How do I
manage this through your site? What concerns should I have?

Why freak out? Take the cash!
Any contacts you get through CL you are on your own with, that's a completely separate entity. If you do sell the car through any other site or offline, just cancel your ad here. You only pay here if you sell it from here and buyer goes through CG's payment process. (BTW I don't work for this site at all, just trying to help you with your question.) You should also be wary of replies from CL ads, LOTS of scammers.
Just seems very odd that I would get four responses, right away, that said they have cash and want to see my car today. One said, sight unseen, wanted to pay the full price in cash and he is over 100 miles away.
Maybe you under priced the car? Have the buyers come to you. Tell them you will meet them in front of a police station or perhaps at your bank. Have the bank check the bills before signing over the title.
As long as they have cash, and you have a safe place to meet. Agree to meet at your bank or a police station or any advertised "safe harbors". Then go from there. Word of caution, one CL scam is the buyer will pay for an item, sight unseen, but wants to pay with a check. Then they say they will pay you extra, provided you wire the extra amount to their shipper. The check bounces and you are out the amount you wired off.
Cashiers checks are easily faked. Any check will take 2 weeks to clear IF it ever does.
