Miss Fire in overdrive

My 2000 dodge 2500 with 5.9 360 has a
misfire only when it shifts into overdrive.
Put new plugs in it new distributer cap and
roter button and it still does it. Like I said
only in overdrive

How do you know it is a miss-fire? Does the check engine light illuminate? If so what are the trouble code(s)? It may be a transmission problem.

It don't show codes now that I've put new injector that was bad in it and put new distributer cap on it. It jumps and jerks and has a backfire when it shifts into overdrive. I was wondering if throttle positioning sensor or maybe plug wires or maybe even fuel filters. Idk it's got me stumped.
