06 Sienna CE cylinder 6 misfiring

18 months ago replaced spark plugs.
Engine light came on, pulled codes. Cyl6
misfire. Problem solved. Today, same
thing. Light, codes, cyl6 misfire. Need coil
packs? Can I just replace #6? It's at the
front acc to the diagram I found.

A quick check would be to swap coil #4 and #6 to see if misfire changes locations. If so it is a coil. If not you need to look at injector and harnesses on injector and coil next.
Only use Denso parts. I have had Toyotas all my life and they do not like other parts. Densofindmypart.com is a good site to find your part numbers for your year and model.
Without look it up, i think it goes like this looking at engine from outside front of car: 1. 3. 5. 2. 4. 6. 1 being on left back by firewall and 6 being in front right closest to you
