I have an '04 Sienna which is the same model as you have so I can go through all the things I've experienced: Power sliding doors should open/shut without any popping/cracking (inspect cables under side windows). Rear power door should open and shut automatically. Try out the sun roof and moon roof. Be aware as to whether the timing belt and/or water pump is due to be changed as that is an expensive repair. Visually inspect the radiator for leaks, they do deteriorate, I had to replace mine. The struts should have been replaced and if you hear vibration in the tires (which could sound like road noise) it could be from strut damage--look for strut wear on the tires. Shifting should be smooth or you could have transmission issues but these Siennas have pretty good transmissions. The right headlight gets water in the lens from a manufacturing defect, make sure there is none in the one you are looking at. Store the rear (fold into the floor) seats and make sure you can unlatch them to pull them back up, those latching systems can wear and bend and leave you stuck. Try the CD changer if it has one (load all 6 discs) and verify the DVD player works if it has that as well. If you toggle the recycle air button on the AC system you shouldn't hear a clicking/popping noise behind the glove box, which would indicate a louver solenoid gear failure (not a deal breaker but maybe you can talk them down if it has this issue). The power steering rack can leak after a while, something to have a mechanic look at. There shouldn't be any stiff points in the steering, if there are there is a U-joint in the steering shaft system that gets corroded on the outer side of the firewall, something also to watch for. Those are some things I'd check for.
