Would a low inflation spare tire cause the sensor lite to stay ON the dash?

if it is a full size spare, yes it also has a tire pressure sensor.

I've seen it happen with a donut spare too, this was on a 2004 Buick. Poor old lady had her tires inflated to 50psi before she decided it wasnt her tires.

Not on topic to your question, just reminds me of the old VW's that used the air from the spare tire, in front, to push windshield cleaner fluid

Yes, you need to have the spare checked whenever you take your Lexus in for service, or do oil changes, to be sure it's always inflated properly. All five tires are linked to the sensor.
