Front Passenger door won't unlock with the key or manually from inside the
car.

Does the fob unlock it? The Focus has a weak inner door handle that breaks free from the sheathed locking cable. The inner and outer handles have their own cables running back to the lock mechanism, as does the lock actuator. You may need to remove the door panel to see what is going on, but before doing that try a little graphite lubricant on the lock/latch area.

GuruB4TJJ answered about a minute ago Fob won't unlock the door but you can feel slight movement on the unlocking part of handle when using the fob all parts of the handle move also can't see any way of removing the door panel while the door is shut & locked

The door does have to be open to remove the panel. I assume you tried all avenues to open the door, inside and outside handles and fob. It seems highly unusual that BOTH handles and the actuator should fail. Some people with other models have had luck with a door that was stuck shut and locked by resetting the fob and actuators. The keyless remote reset is "turn ignition from OFF to RUN 4 times within 6 seconds. Within 20 seconds, press any button on remote. Within 20 seconds press any button on second remote." IDK if there is another procedure for FOB or lock actuators or not. The fact you can feel movement in a door handle when pressing the FOB button seems to indicate the actuator is working, but not affecting the lock mechanism. Try looking at the gaps along the door, between the door itself and the rest of the body. If it appears the door is sagging even a little, it may be binding the locking mechanism on the jamb latch. If so, use a pry tool wrapped in cloth (to prevent scratches in the paint) to gently lift the door enough to take the pressure off the latch and then try the FOB. If the door handles are truly broken, chances are they broke right at the handle, where the cable connects to it. If so, you may be able to use needle nose pliers to grab the end of that cable and yank it. That should open the lock. That's about the extent of DIY repair tips, if none of these work you may want to contact the dealership for suggestions, or AAA may be able to get the door open.

I have a 2010 Ford Focus. The right rear passenger door would not open from the outside, inside handles, the key fob or by using the actuator. I recalled that earlier in the morning when I had dropped by husband off to work(car running) the door had opened as usual by using the actuator. I started the car and used the actuator, sure enough, it worked, even better the door appears to be working as normal. I did lubricate the latch with WD-40 after the door was open.
