check the fuse for this. it could also be the switch itself.

If you don't have an owners manual, you can Google the location of the fuse box for your car. On many vehicles it is on the drivers side of the dash accessed with the door open. On some vehicles it is under the hood or in the glove compartment. There may even be more than one. Your owners manual should be able to help. You can Google almost anything these days, but if you want an owners manual, try books4cars.com.
Mine was doing same thing , gasket around the door was loose and was not in place , fixing gasket took care of it .
I have a 2013 rx360 the back door will not close and latch. It will go down but come right back up
My hatchback controls don’t open automatically
I have a 2017 Lexus Rx350 and my trunk would not close with the key fob would not close with interior button. I searched everywhere with no luck. I removed my battery twice and no luck. I tried manually closing and it wouldn't budge at first. After battery reset it takes a few minutes for the key fob to work again only in the up position. This is what worked for me.. So I manually started closing the trunk little by little if you try to go all the way down it will lock up and not budge. Push down a little stop pull up a tiny bit then continue to do that till it is fully closed. Once the latch grabs and lock it resets the motor i guess. I opened with the key fob and closes with interior button and trunk button. Hope this helps someone i spent a bout 2 hrs resetting battery and checking all the fuses. I thought for sure I was heading to the dealer to get fixed.

I took my Lexus RX 350 in yesterday for this reason, the lift gate button was not working, it would only beep so I would have to manually pull it up or down. 130$ later the mechanic says that there is a button in the glove compartment that might have been pushed that shuts that mechanism off for reasons I can't remember. I was too focused on how much they charged me to remember that detail
