possible misfire

i have a 2004 dodge 2500 5.7 hemi, taking off in the truck is fine and when i get up to crusin speed and give it a little more gas it wants to misfire but when i thrash the throttle it clears up. would anyone know whats going on? i replaced the fuel regulator and still didnt fix it...

How many miles? Has it ever been tuned? It sounds like a bad plug boot, the fact that it will go wide open eliminates the fuel system.
