I stopped driving my 2000 E320 Mercedes Benz in Feb 2013 due to shortage in brake lights/tail lights. I have not drove the car since, it has been sitting in driveway. I started the car to move to back of driveway and it would not move/ found a major transmission leak. What would have caused this leak it wasn't leaking before I parked it and didn't drive it anymore?

I would say you had some seals dry up and that's what is leaking.
