how many miles can I expect my 2001 Lexus ES to last?

All depends on how well it was maintained. I currently have a Ford with over 300K on it, no major work, still on original tranny and torque converter. Your Lexus if it was maintained should match that, but then sometimes factory components even when maintained can die an early death.
